Aim and content
This course is free of charge for PhD students at Danish universities (except Copenhagen Business School). All other participants must pay the course fee.
Anyone can apply for the course, but if you are not a PhD student, you will be placed on the waiting list for the course until enrollment deadline. After the deadline of enrollment, available seats will be allocated to students on the waiting list.
Learning objectives
A student who has met the objectives of the course will be able to:
1. Have obtained a basic knowledge of the underlying principles, mechanism of action and the potential as well as the limitations of genome editing using CRISPR/Cas9.
2. Be able to design target specific CRISPR/Cas9 reagents and incorporate their use in own research project.
3. Be able to analyse and assess own results as well as those obtained by others using CRISPR/Cas9 and be prepared for the upcoming developments in the field of genome editing.
Content
This theoretical course will provide an overview of the principles and use of the CRISPR/Cas9 system for genome editing, from the basics to the most advanced applications. Participants will learn how to design CRISPR/Cas9 reagents for a desired genomic target, select appropriate delivery vectors and editing strategy for a given project, principles for generation of gene knockout or knockin-modified/base-edited cells and become familiar with the various methods for mutation detection using the current technologies. Genome-wide CRISPR screening, CRISPRi, CRISPR paint and other CRISPR applications will be introduced. Advanced research stories using CRISPR/Cas9 will be presented.
The course includes the one-day mini-symposium on “Precise Genome Editing using CRISPR/Cas9” featuring international speakers form leading laboratories
Design of CRISPR/Cas9 reagents (gRNAs) using online tools; delivery methods (transfection/transduction) and associated vector systems; basic rules for knockout and knockin, including design of knockin targeting constructs; enrichment strategies for genome edited cells (FACS, antibiotics selection); methods for detection of CRSPR/Cas9-induced knockout/knockin/base editing mutation and clone validation; off-target considerations and rescue strategies; Alternative genome editing tools (TALENs/ZFNs) and CRISPR applications (genome-wide screening/CRISPRi/CRISPRa/CRISPR paint etc), animal engineering; examples of research applications of CRISPR/Cas9 technology.
Participants
The course is intended for PhD students who have a desire to use or are already applying CRISPR/Cas9-based genome editing in their research. No previous experience in genome editing is required. The course has room for 25 participants.
